Drug Interactions

Propoxyphene is mainly composed of the cytochrome P450 3A4 isoenzyme system (CYP3A4), so potential interactions may occur when propoxyphene is administered simultaneously with agents that affect CYP3A4 activity.

Propoxyphene metabolism can be altered by potent CYP3A4 inhibitors (such as ritonavir, ketoconazole, itraconazole, troleandomycin, clarithromycin, nelfinavir, nefazodone, amiodarone, amprenavir, aprepitant, diltiazem, erythromycin. CYP3A4 may reduce the activity of propoxyphene. Chemical inducers of CYP3A4 like rifampin can lead to improved metabolite levels.
